Antioxidant and cytotoxic activity of methanol extract of cocoa pod husk (Theobroma cacao L.) has been tested using 1,1-diphenyl-2-pycrixlhidrazil (DPPH) and the Brine Shrimp Lethality Test (BSLT) method. The results of the antioxidant activity test of TCM and TCA extracts are 135.60 and 107.94 ppm and the cytotoxic are 123.02 and 151.35 ppm. The cocoa pod husk is then extracted using methanol, and the extract is partitioned using acetone. Furthermore, its chemical components were seperated using column chromatography with a stationary phase of silica gel and a mobile phase using an eluent system, resulting in 11 combined fractions (TCA1-TCA11) with successive IC50 values 308,97; 258,34; 114,24; 18,00; 22,79; 7,37; 59,59; 215,30; 124,19; 60,54; 98,67; and 2,41 ppm. In addition, the IC50 value was determined successively: 100; 10,96; 436,51; 1174,89; 51,28; 38,81; 147,91; 26,30; 1096,47; 269,15; and 26,30 ppm. The TCA6 fraction showed the highest antioxidant activity with an IC50 value of 7.37 ppm, while the TCA fraction 2 showed the highest cytotoxic activity with an IC50 value of 10.96 ppm. © 2024 Author(s).
